In the first part of the current thesis the aldol-Tishchenko reaction of ketone aldols as enol equivalents has been developed as an efficient strategy to furnish differentiated 1,3-anti diol monoesters in one step. The thermodynamically unstable ketone aldols undergo a facile retro-aldolization to yield a presumed zirconium enolate in situ which undergoes the aldol-Tishchenko reaction in typically high yields and with complete 1,3-anti-diastereocontrol. Evaluation of a broad range of metal alkoxides as catalysts and optimization of the reaction protocol led to a modified zirconium alkoxide catalyst with attenuated Lewis acidity and dichloromethane as solvent which resulted in suppression of the undesired acyl migration to a large extent. Various ketone aldols have been prepared and subjected to the general process giving rise to a broad range of differently substituted 1,3-anti diolmonoesters which may be hydrolyzed to the corresponding 1,3-anti-diols. In the second part an enantioselective, catalytic version of the title reaction was developed employing chiral TADDOLs, BINOLs and N-oxides as ligands attached to zirconium. The 1,3-anti diolmonoesters were obtained in enantioselectivities up to 60% ee.

Im ersten Teil der vorliegenden Doktorarbeit wurde die Aldol-Tishchenko-Reaktion von Keton-Aldolen als Enolat-Äquivalente als effiziente Strategie zur Synthese von 1,3-anti-Diolmonoestern in einem Schritt entwickelt. Die thermodynamisch instabilen Keton-Aldole bilden nach einer Retro-Aldol-Reaktion in situ ein Zirconium-Enolat, welches nachfolgend die Aldol-Tishchenko-Reaktion in typischerweise hohen Ausbeuten und vollständiger 1,3-anti-Diastereoselektivität eingeht. Der Einsatz verschiedener Metallalkoxide als Katalysatoren und die Optimierung der Versuchsbedingungen lieferte ein modifiziertes Zirconiumalkoxid mit verminderter Lewis-Basizität als Katalysator sowie Dichlormethan als Lösungsmittel, wodurch die unerwünschte Acylgruppenwanderung zu einem großen Teil unterdrückt wurde. Verschiedene Keton-Aldole wurden hergestellt und unter den optimierten Bedingungen umgesetzt, wodurch eine Reihe unterschiedlich substituierter 1,3-anti-Diolmonoester erhalten wurde, die zu den entsprechenden 1,3-anti-Diolen hydrolysiert werden konnten. Im zweiten Teil wurde eine enantioselektive, katalytische Variante dieses Verfahrens entwickelt, indem chirale TADDOLe, BINOLe und N-Oxide als Liganden für Zirconium verwendet wurden. Dabei wurden die 1,3-anti-Diolmonoester mit Enantioselektivitäten bis zu 60% ee erhalten.
